Developed by researchers at UFMG, this therapeutic vaccine stimulates the production of antibodies that prevent cocaine from reaching the brain; the technology has already been patented in Brazil and the United States.

A vaccine developed by researchers at the Federal University of Minas Gerais (UFMG) could open a new frontier in the treatment of cocaine and crack addiction. Named Calixcoca, the technology stimulates the immune system to produce antibodies capable of preventing cocaine from reaching the brain, thereby reducing its effects and potentially helping patients avoid relapse during the recovery process.

The research is led by Professor Frederico Duarte Garcia of the UFMG School of Medicine and has been in development for over a decade. The project has already completed preclinical stages—conducted on animals—during which researchers observed antibody production and the blocking of the drug's passage into the central nervous system.

This approach distinguishes Calixcoca from traditional medications because the vaccine does not act directly on the brain. Instead, it mobilizes the body's own immune system to prevent cocaine from reaching the central nervous system.

How Calixcoca works

The technology utilizes a molecule known as V4N2—a calixarene—capable of inducing the body to produce antibodies against cocaine. When an immunized person comes into contact with the drug, these antibodies bind to the cocaine molecules while they are still in the bloodstream. The binding forms a molecular complex too large to cross the blood-brain barrier, the structure that controls the passage of substances from the blood to the brain.

As a result, the amount of cocaine capable of reaching the central nervous system is reduced, and consequently, the effects caused by the drug can also be blocked or diminished.

Preclinical experiments conducted by the UFMG team confirmed antibody formation and the blocking of cocaine's passage into the central nervous system in animal models.

This strategy is particularly significant because cocaine's effect on the brain is directly linked to the intense sensation of reward that fuels the cycle of consumption and addiction.

Vaccine does not prevent initial exposure to the drug

Despite its name, Calixcoca should not be viewed as a conventional preventive vaccine, such as those used against infectious diseases.

Its purpose is therapeutic.

The researchers propose using the vaccine primarily as a complementary tool in treating individuals who have already developed an addiction and wish to maintain abstinence.

The goal is to create a biological barrier against the drug's effects, increasing the likelihood that patients can navigate critical treatment periods without re-experiencing cocaine's psychoactive effects.

Frederico Garcia explains that this strategy could provide more time for these individuals to rebuild aspects of their lives that were damaged by addiction. “We demonstrated a reduction in the drug's effects, suggesting efficacy in treating addiction. We envision using the drug to prevent relapse in patients undergoing treatment, giving them more time to rebuild their lives without the drug,” the researcher stated.

This means that Calixcoca is not intended to replace medical, psychological, and social support. If its efficacy is subsequently proven in humans, the vaccine could become part of a broader set of strategies used during recovery.

A gap in addiction treatment
The development of Calixcoca addresses a challenge that has engaged researchers from various countries for years: the lack of a specific, established pharmacological treatment for cocaine addiction.

Currently, behavioral and psychosocial interventions remain the primary treatment strategies. There is no medication specifically approved by the U.S. regulatory agency for cocaine use disorder.

In this context, a technology capable of acting biologically to prevent the drug from producing its effects could add a new tool to existing approaches.

However, the researchers themselves emphasize the need for caution. Positive results in animals do not automatically guarantee the same outcome in humans. Safety, dosage, immune response, potential adverse effects, and efficacy will need to be evaluated across the various stages of clinical trials.

Patents in Brazil and the United States

One of the project's key recent milestones was the granting of patents for Calixcoca.

The technology is patented by the National Institute of Industrial Property (INPI) in Brazil and also in the United States. UFMG and the Minas Gerais State Research Support Foundation (Fapemig) are co-holders of the patent.

In addition to intellectual property protection, the project has received a new financial boost.

The Government of Minas Gerais announced an investment of approximately R$ 18.8 million dedicated to the development of Calixcoca and the steps required to eventually enable its clinical evaluation.

According to UFMG, the total funding amounts to R$ 18.84 million, distributed in installments over several years. Additional resources have also been allocated to the project through Fapemig public calls for proposals.

The path to human trials

Despite these advances, Calixcoca remains an experimental product and is not yet available for public use.

Before reaching the clinical trial stage, researchers must still complete further evaluations and generate evidence under certified conditions that meet regulatory requirements.

In an update released by UFMG, researchers explained that additional testing is required before moving to human trials. The university projects a timeframe of two to four years for this transition, depending on the results obtained during the process.

Once authorized, Phase 1 will aim primarily to analyze the product's safety in humans and identify potential adverse effects.

Only after progressing through the various stages of clinical research will it be possible to determine whether Calixcoca truly works safely and effectively in patients. Brazilian research gains international recognition

Calixcoca has also garnered recognition beyond the laboratory.

The project won the Euro Health Innovation Award in the Technological Innovation Applied to Health category. The award provided €500,000—approximately R$ 2.6 million at the time—for the technology's development.

The initiative brings together a multidisciplinary team of researchers from fields such as medicine, pharmacy, and chemistry.

In addition to Frederico Duarte Garcia, the project involves researchers such as Maila de Castro, Gisele Goulart, and Ângelo de Fátima, as well as members affiliated with the Research Center on Vulnerability and Health (NAVeS). The Research Development Foundation (Fundep) supports the project by assisting with the management and facilitation of processes required for the research's development.

The project also receives support from institutions such as Fapemig, Capes, CNPq, Finep, the National Secretariat for Drug Policies, and the Ministry of Science, Technology, and Innovation.

A promising development that still faces a crucial test

The progress made with Calixcoca represents a significant prospect for a field that still faces major therapeutic limitations.

By using antibodies to try to prevent cocaine from reaching the brain, the technology proposes an approach that differs from those currently used to treat substance dependence.

However, the path from positive results in animal studies to a medication available in healthcare services still involves several stages.

Human studies will be essential to determine whether the mechanism observed in preclinical experiments can offer safety and therapeutic benefits to patients.

If the results are positive, Calixcoca could become an important complementary tool for people struggling to maintain abstinence and rebuild their lives free from cocaine and crack—placing Brazilian research at the forefront of the search for new strategies against substance dependence.
