The Brazilian Registry of Clinical Trials (ReBEC) is a free access virtual platform for recording experimental and non-experimental studies carried out on human beings and conducted in Brazilian territory, by Brazilian and foreign researchers. ReBEC is a joint project of the Ministry of Health (DECIT/MS), the Pan American Health Organization (PAHO/OPAS) and the Oswaldo Cruz Foundation (FIOCRUZ). ReBEC's Executive Committee is composed of the aforementioned institutions and the National Health Surveillance Agency (ANVISA). ReBEC is an important source of information for patients, healthcare professionals, researchers, companies, and research centers. The study aims to gather initial study findings by the end of 2026 while further data is expected during 2027.

Skylark Bio has dosed the first patient in its SONIX Phase I/II clinical trial assessing SKY-GJB2 as a potential gene therapy for children with hearing loss linked to GJB2 gene changes.

The trial is designed to assess the safety, tolerability, pharmacokinetics and pharmacodynamics.

It will also evaluate the preliminary efficacy of SKY-GJB2’s single unilateral intracochlear injection in paediatric patients aged between nine months and seven years across multiple sites.

The study aims to gather initial study findings by the end of 2026, while further data is expected during 2027.

SKY-GJB2 is designed to deliver a functional copy of the GJB2 gene directly to supporting cells within the cochlea that are integral for the natural expression of connexin 26, a protein required for normal hearing function.

Mutations in the GJB2 gene disrupt this protein’s function and impact the gap-junction network within the inner ear, which is crucial for hearing.

Artificial intelligence isn't just speeding up early-stage drug development, it's starting to unlock millions of dollars' worth of new efficiencies in clinical trials on cancer treatments, research shared first with Axios shows.

Why it matters: AI-powered tools could shave months off time-consuming processes like recruiting and enrolling patients, monitoring results and interpreting data.

• That could free up resources for more clinical studies and possibly lower the high failure rate for new drugs.

What they found: A new Tufts Center for the Study of Drug Development analysis found AI agents can accelerate clinical development of cancer drugs by approximately 10 weeks and reduce direct operating costs by as much as $5.6 million in late-stage trials.

• The efficiencies increase with the number of tumors a drug can target: An experimental treatment with 50 active uses could see net benefits of as much as $565 million, Tufts found.
• Tufts applied a clinical monitoring agent from Medable, which provides a platform to support clinical trials, to an unspecified oncology drug development program in phase 2 and 3 trials.

What they're saying: Ken Getz, the Tufts center's executive director, said deploying the agent led to new efficiencies, such as reducing the number of on-site visits, accelerating enrollment in the trial and locking in the data.

• "To our knowledge, this is the first time that [predictive] modeling based on actual use and benchmark data has been applied to quantify the net financial impact of an agentic AI solution in a drug development program," he said.

Computer simulations can help to improve the design of clinical trials during the planning phase. This is the conclusion reached by an international team of experts led by the Medical University of Vienna in a research article published in "Nature Reviews Drug Discovery". The authors demonstrate how simulations can be used to systematically test, compare and refine different trial designs in advance. At the same time, they set out recommendations on how such simulations should be conducted in a robust and transparent manner.

When you think of a clinical trial, a few scenarios may come to mind. Maybe you picture agreeing to take a medication that could help your specific condition. You think of signing a consent form that commits you fully until the study has ended, with no way to leave. 

There are many common misconceptions about clinical trials, but potential participants should know that informed consent is the backbone of these studies. 

In reality, clinical trials are founded on the idea of participant safety and consent. During the initial stages of a trial, potential participants are allowed to review informed consent, which helps them make an informed decision — not in any way take away their choices. Informed consent is an ongoing communication process designed to protect participants — not simply a form they must sign.

What Is Informed Consent?
Informed consent is the process by which the research team explains the study so you can decide whether participation is right for you. As a potential volunteer, you will receive a full explanation of all aspects of the study. 

Edward S. Kim, MD, MBA, serves as physician-in-chief at the City of Hope National Medical Center and system director for clinical trials. He spoke on a trial’s priorities from a researcher’s perspective.

Dr Olivier Kasita is a physician working at the Elikya Ebola treatment centre in Bunia, Democratic Republic of Congo. This centre has been supported by Médecins Sans Frontières (MSF) since the beginning of the Ebola disease outbreak. Through his work at the centre, he has been directly involved in the care of patients throughout the response. Dr Kasita answers questions about the new PARTNERS clinical trial.

What do clinical trials involve? How do they work?
The PARTNERS trial is the World Health Organization (WHO) sponsored international clinical research trial conducted through close collaboration between the national health authorities, research institutions, outbreak response partners, and international organisations. It aims to rapidly generate high-quality evidence on the safety and efficacy of two promising treatments, MBP134 et Remdesivir, for patients who tested positive for Bundibugyo virus disease.

Participants who volunteer to join the trial are randomly assigned to one of four study groups: one receives both MBP134 and Remdesivir, one receives MBP134 alone, one receives Remdesivir alone, and one receives the current standard of care without either investigational treatment. This randomised design allows researchers to compare outcomes across groups and determine whether the treatments provide a meaningful benefit.

This study gives patients a chance to receive one of the treatments that researchers believe may work better than the current standard.
