The US is driving innovation in the peptide therapeutics space, especially around GLP-1s.

Investment in peptide therapies is set to soar in the next decade. A recent financial report from Grand View Research predicts a market value of almost $300 billion by 2033, more than double their 2025 evaluation of $140 billion. The US currently dominates the peptide space at more than 60% of the market share. 

What appears to be driving this growth is the stratospheric success of glucagon-like peptides (GLP-1s). These have fueled clinical trial investments to discover additional indications for GLP-1s while spotlighting the abilities of other peptide drugs. 

From uses in oncology to the treatment of rare diseases, peptide popularity is attracting significant investor attention.

More than weight loss
Much of the focus on GLP-1s has been on weight loss, with the World Health Organization (WHO) releasing guidelines recommending their use for the “long-term management” of obesity. While this will certainly generate significant capital within the peptide therapies market, multiple studies have highlighted that GLP-1s aren’t just a one-trick pony.
A landmark study published in the Journal of the American Medical Association (JAMA) evaluated data from over 58,000 patients with type II diabetes, heart failure, and obesity. The study found a 42% risk reduction of death or hospitalization from heart failure in patients treated with GLP-1s compared to the placebo group. What’s more, there was nearly double the incidence of hospitalization and death in patients treated with placebos compared to those treated with GLP-1s.

But the benefits of GLP-1s don’t stop at risk reductions in heart failure. A Phase IIb clinical trial (NCT01843075) of over 200 patients published in Nature Medicine has shown the benefits of liraglutide for slowing disease progression in Alzheimer’s. In the double-blind randomized controlled trial, researchers found that liraglutide-treated patients had significantly less shrinkage of tissue in the temporal, parietal and frontoparietal lobes. Alzheimer’s patients on liraglutide also showed significantly slower progression of disease compared to the control group.

GLP-1s have more promise in multiple disease areas than initially thought, setting the stage for increased investment and applications (as long as synthesis bottlenecks can be overcome).

Cleverer than chemo
The oncology treatment landscape has been transformed over the last decade by several innovations, chief among them targeted immunotherapies. Preclinical research now suggests that oncolytic peptides and peptide conjugates could be an invaluable asset in this arena.
A recent study of ruxotemitide has shown promise in preventing metastasis in melanoma patients. The peptide therapeutic works by turning “cold” tumors “hot” – that is, making them visible to the immune system. Ruxotemitide is injected into the tumor site to incite immunogenic cell death, which reveals the cancer for detection. The data so far is promising: in vivo animal models showed both tumor shrinkage and long-term immune memory against tumor antigens.

Peptide conjugates have also shown significant promise against an aggressive form of breast cancer. An animal study published in NAR Cancer describes an oligo-peptide conjugate that can target and silence overexpressed microRNAs in triple-negative breast cancer. The team achieved this by binding a microRNA inhibitor to an IGF-1 like peptide, which allows the conjugate to be actively transported into cancer cells. 

After just two weeks, the peptide conjugate not only halted tumor progression, but shrank mean tumor mass by 74% in mice without inducing end-organ toxicity.  

Delivering the future
As science advances, the therapeutic potentials of gene therapy become increasingly impressive, with the ability to provide rescue from rare disease. But reliance on viral vectors can come with risks, as several widely publicized clinical trial fatalities have shown. Researchers are therefore scrambling to find safe nonviral methods of gene therapy delivery. Once again, peptides may hold the answer.
Results published in Nature Communications suggest that attaching a nuclear localization signal (NLS) peptide can help direct DNA to the nucleus. This would allow several benefits over other conjugate approaches like nanobodies, including smaller size, as well as a more streamlined mechanism of entry into the nucleus. The research team was able to increase gene expression up to 18-fold using their NLS peptide conjugate when compared with a control.

Peptide therapies are testing their mettle against popular alternatives like antibodies and holding their own. Clearly, investors are taking notice. A compound annual growth rate (CAGR) estimated at a minimum of 8% per annum indicates peptides are only set to get more popular in the coming years.
