This is a cross-sectional, observational study without intervention. A total of 42 patients meeting the eligibility criteria will be divided into two experimental groups based on the absence or presence of type 2 diabetes mellitus (T2DM): i) PE group (patients with periodontitis; n=21); and PE/DM group (patients with periodontitis and T2DM; n=21). Patients will undergo a full-mouth periodontal examination, anthropometric assessment, and blood sample collection for laboratory analysis. Subgingival biofilm and fecal samples will be collected for analysis via sequencing of the 16S rRNA gene. Questionnaires will be administered to gather data on sociodemographic parameters, lifestyle, and dietary and bowel habits.

Trata-se de um estudo observacional, de temporalidade transversal, sem intervenção. Um total de 42 pacientes que preencherem os critérios de elegibilidade será dividido em dois grupos experimentais, de acordo com a ausência ou presença de diabetes mellitus tipo 2 (DM2): i) grupo PE (pacientes com periodontite; n=21); grupo PE/DM (pacientes com periodontite e DM2; n=21). Os pacientes serão submetidos ao exame periodontal de boca toda, à avaliação antropométrica e à coleta de amostras de sangue para realização de exames laboratoriais. Amostras de biofilme subgengival e de fezes serão coletadas para análise por meio de sequenciamento do gene 16S rRNA. Serão aplicados questionários para levantamento de parâmetros sociodemográficos e de estilo de vida, hábitos alimentares e evacuatórios.

Inclusion criteria:

Volunteers of both genders; minimum age of 18; presence of at least 15 natural teeth, excluding third molars and teeth indicated for Extraction; diagnosis of generalized Periodontitis, stages III or IV, grades B or C; 30% or more of teeth with probing depth and clinical attachment level greater than or equal to 5 millimeters (mm), with bleeding on probing; patients with Type 2 Diabetes Mellitus must have had the diagnosis for at least 5 years, with a glycated hemoglobin level between 7% and 11% and treatment with Insulin or Hypoglycemic Agents

Long-term use of Anti-inflammatory or Immunosuppressive medication; Periodontal Treatment and/or use of Antimicrobials or Probiotics within the last 6 months; continuous use of mouthwashes containing Antimicrobial Agents within the last 6 months; extensive prosthetic rehabilitations; current smoker or smoker within the last 5 years; Pregnancy; Lactation

Primary outcomes:

Differences in fecal microbiota diversity between the experimental groups are expected. This outcome will be evaluated through alpha diversity analysis using the Shannon index. The outcome will be verified by comparing the Shannon index values between the experimental groups.
